Rhomboid intramembrane proteases are widely conserved and regulate a number of cellular processes, including medically relevant ones. Their inhibitors useable in cell biological experiments have been lacking until my group discovered and developed peptidyl ketoamides as potent rhomboid inhibitors (Ticha et al. Cell Chem Biol 2017). We develop this compound class further, to refine their design principles and develop tools for selected rhomboid proteases (Polachova et al. J Med Chem 2023; Gandhi et al. Cell Chem Biol 2020).
The project will entail recombinant membrane protein expression and purification, cell-free translation, enzyme kinetics, basic cell biology and can entail synthetic chemistry and/or x-ray crystallography. The ideal candidate would show experience in biochemistry and molecular biology. Exposure or experience in membrane proteins, enzyme kinetics or structural biology would be a bonus. The candidate would overlap with an experienced PhD student and would collaborate with a staff scientist chemist, and other postdocs and PhD students.
