Monitoring of fertility parameters of sperm and testicular tissue in patients with testicular cancer with respect to their therapy and development of evidence-based methodology for Centres of Assisted Reproduction
This project aims to address Testicular germ cell tumours (TGCT)-related sperm and testis germ cell pathologies from both the clinical and experimental angle. Its goal is to deliver novel diagnostic strategies for the evaluation of samples from TGCT patients prior to surgery and after the therapeutic intervention, in order to be able to monitor and compare the quality of fertility parameters and to recommend which cryopreserved semen should be used in programmes of assisted reproduction. Thus, the outputs of the project should considerably improve the patient´s chances of successful conception.
The Postdoctoral Fellow will mainly be responsible for genomic and proteomic methods involved in the project, working with human male gamete and tissue biopsies under the obtained Ethical Consent. The project involves sperm motility monitoring by the Computer Assisted Sperm Analyser (CASA), cytometric analyses of early and late stages of apoptosis, acrosome integrity evaluation and assessment of epigenetic parameters and detailed evaluation of mitochondrial activity, characterisation of sperm mitochondrial function and the resulting ratio of ATP generated by OHPHOS and glycolysis. The methods will also include IF, IHC, WB, IP, confocal and super-resolution microscopy, antibodies selection for specific diagnostic kit requirements, cell cultures experiments, gene expression, PCR, qRT-PCR; Further data analysis, preparation for the manuscript and active part in writing publications and presenting the data is required.

We are looking for a motivated bright Postdoctoral Fellow with background in reproductive biology, holding Ph.D. degree in molecular and cell biology, developmental biology or reproductive biology. The scientific focus on spermatogenesis, sperm maturation, sperm evaluation, gamete handling, sperm-egg interaction and embryology is a strong advantage. The knowledge should cover genomic and proteomic method of wide range. The candidate should be able and prepared to work with human gametes and tissues. Excellent English is required. The candidate should be a team-player and willing to work with other lab members and international collaborators.
