Neurosteroids are endogenous compounds that play a crucial role in various physiological and pathological conditions of the brain and nervous system. Therefore, neurosteroids have been considered a potential solution for the treatment of many disorders of the central nervous system. However, the rate-limiting step in the development of novel compounds for neuroscience-related research is their blood-brain-barrier permeability. Hundreds of steroids have been biosynthesized in the human body and many of them are neurosteroids. The structure-activity background for their ability to permeate through the blood-brain barrier is not clear. The proposed project will focus on the characterization of the permeability of endogenous compounds, their modifications, and the follow-up synthesis of novel compounds from the perspective of their ability to permeate for potential use as novel drug-like compounds for the central nervous system.
