Project TESCHCALE: The primary focus of the TECHSCALE project (https://www.techscale.cz/en/) is the design, implementation, characterization, and application of a new class of materials prepared by precision single-atom engineering based on advanced nanotechnologies. During the project implementation, we expect fundamental discoveries that will push ahead the frontiers of current nanotechnologies. Furthermore, we aim at societally important applications in the field of physical and chemical sciences, namely diagnostics, theranostics, chemical catalysis, and energy storage. Adopting a completely new strategy in this field, we will include, from the beginning, as an integral part of the project, also the assessment of societal impacts and public acceptance of new technologies, including the legal framework and regulatory aspects in accordance with the rules of responsible approach to research and innovation and the objectives of sustainable development. The applicant responsibilities: The successful candidates will join the research groups at CATRIN and work on TECHSCALE project related to the technologies beyond nanoscale. The candidate will receive a two-year contract at Palacký University Olomouc in the Czech Republic, which could be extended up to five-years in case of positive annual evaluation.   • carry out the excellent basic research in the respective field;

Patescibacteria’ is a phylum described only in 2015 and represents one of the biggest discoveries in microbiology in the last decade. It is believed to encompass between ~15% and 26% of bacterial diversity, but only a few cultures are available to date. These few representatives were found to be obligate episymbionts on other prokaryotes. Most of what we know about ‘Ca. Patescibacteria’ is derived from genomic data, but in general CPRs have extremely unusual characteristics, such as a very limited number of metabolic pathways, reduced genomes and cell sizes, and peculiar ribosomes that lack proteins which were considered essential for survival. Many genomes were found to have minimal amino acid and cofactor biosynthesis pathways, and till now no CPR has been identified to produce membrane phospholipids, which adds to the enormous intrigue regarding their lifestyle strategies. In the current project, we aim to provide unique information about CPRs temporal variability, functional potential, and ecological role in freshwater lakes. Additionally, we will approach one of the most difficult aspects in need of clarification – the identity and range of CPR hosts. Our final goal is to bring new CPRs into culture using reverse genomics and study their behavior and biology in detail. REQUIREMENTS:
  • Applicants should hold a M.Sc. in Biological Sciences (e.g., Limnology, Microbiology, Ecology).
  • Candidates must be strongly motivated to work in an eco-genomics laboratory.

Patescibacteria’ is a phylum described only in 2015 and represents one of the biggest discoveries in microbiology in the last decade. It is believed to encompass between ~15% and 26% of bacterial diversity, but only a few cultures are available to date. These few representatives were found to be obligate episymbionts on other prokaryotes. Most of what we know about ‘Ca. Patescibacteria’ is derived from genomic data, but in general CPRs have extremely unusual characteristics, such as a very limited number of metabolic pathways, reduced genomes and cell sizes, and peculiar ribosomes that lack proteins which were considered essential for survival. Many genomes were found to have minimal amino acid and cofactor biosynthesis pathways, and till now no CPR has been identified to produce membrane phospholipids, which adds to the enormous intrigue regarding their lifestyle strategies. In the current project, we aim to provide unique information about CPRs temporal variability, functional potential, and ecological role in freshwater lakes. Additionally, we will approach one of the most difficult aspects in need of clarification – the identity and range of CPR hosts. Our final goal is to bring new CPRs into culture using reverse genomics and study their behavior and biology in detail. REQUIREMENTS:
  • Applicants should hold a PhD in Biological Sciences (e.g., Limnology, Microbiology, Ecology).
  • Candidates must be strongly motivated to continue their work in an eco-genomics laboratory.
  • Prior experience in one or more of the following areas is favorably considered:

But we are only beginning to understand the consequences of this eco-evolutionary process for dynamics of host – parasite interactions and stability of entire communities. We ask how rapid evolution impacts maintenance of diversity in communities and maintenance of genetic variation in populations - processes which have been mostly studied separately. To address these questions, our collaborative project uses a novel experimental community model system of wild Drosophila species and their parasitoids from tropical Australia. We are able to perform multigenerational laboratory microcosm experiments and track eco-evolutionary dynamics in fine detail. The candidate will use a combination of the following approaches: laboratory experiments on rapid evolution, and maintenance of species diversity and genetic variation in communities, experimental evolution, population genomics, eco-evolutionary modelling, and field surveys of Drosophila - parasitoid food webs in Australian tropical rainforest. The specific PhD projects will result from a discussion between the candidate and the supervisor. The candidate will take part in the fieldwork in Australia, probably already in the second half of 2024. REQUIREMENTS: A MSc degree is required to enter PhD in Czech Republic. The research activities will include the characterization of thermally induced phase transitions and in-situ real-time experiments during thin films growth. The research will be performed at the Department of Condensed Matter Physics, Masaryk University, at CEITEC Nano Infrastructure (both in Brno) and at synchrotron facilities. The position is part of a recently granted project to find phages of abundant freshwater bacteria (see below), but it can be tailored to the applicant’s interest. Project background: Phages are present in all ecosystems and structure microbial communities through the regulation of host populations and the release of host-bound nutrients through lysis. Metagenomics revealed a huge phage diversity in nature but often failed to assign hosts to phages. Especially in freshwater environments, this is partly due to the lack of cultured reference phages and their hosts. Recent advances in bacterial cultivation enabled the isolation of abundant freshwater bacteria (e.g., Fonsibacter, Planktophila, Methylopumilus). We have established a large freshwater bacteria collection (>7000 isolates) that includes these abundant species. Here we aim to use these bacteria in a high throughput phage isolation approach to isolate new phages and to bona-fide identify phage-host pairs. We will study the host range, natural abundances and dynamics of newly isolated phages. We will completement these approaches with culture independent approaches to quantify isolated phages, identify prophages in the genomes of isolates and in metagenome assembled genomes from long-read metagenomic data of environmental bacteria. The project hence plans to close an important gap in viral ecology by assigning hosts to phages and investigate their natural dynamics in a freshwater reservoir. REQUIREMENTS:
